Viruses that do not express p12 (HTLV-1p12KO) cannot persistently infect macaques, and the transient depletion of monocytes prior to virus exposure allows early seroconversion, which is not sustained over time. To test whether HTLV-1 persistence depends on a monocyte reservoir or if monocyte depletion provides a transient immune evasion benefit, researchers at the National Cancer Institute simultaneously depleted NK, CD8+ T cells, and monocytes (triple depletion) prior to exposure to HTLV-1p12KO. Triple depletion resulted in the exacerbation of infection and the complete rescue of HTLV-1p12KO infectivity. This underscores the role of monocytes in orchestrating anti-viral immunity and the importance of p12 in hijacking host immunity.
